Overview

HSV-1716 is a replication-restricted oncolytic herpes simplex virus developed as a cancer therapeutic. It contains a deletion of the ICP34.5 gene, which is a neurovirulence gene that enables viral replication in neurons. This deletion provides tumor-selective replication properties, allowing the virus to replicate in and kill cancer cells while largely preventing replication in normal cells. The virus works by exploiting the weaker PKR-linked defenses in tumor cells compared to healthy differentiated cells. HSV-1716 has been studied via intratumoral injection, intravenous administration, and intrapleural delivery in various cancer types including glioma, mesothelioma, melanoma, and solid tumors in both adult and pediatric patients[1][2][4][7].
