HSV-SUR15 is an engineered oncolytic herpes simplex virus (oHSV) designed for the treatment of glioblastoma (GBM). It utilizes a tumor-specific survivin promoter to control the expression of the viral neurovirulence gene ICP34.5 and the ribonucleotide reductase gene ICP6, thereby restricting viral replication and cytotoxicity to survivin-expressing tumor cells. In addition to its direct oncolytic activity, HSV-SUR15 is engineered to secrete Interleukin-15 (IL-15), a cytokine that promotes the activation and proliferation of T cells and natural killer (NK) cells. This dual mechanism aims to induce a potent and durable anti-tumor immune response while minimizing off-target neurotoxicity. Preclinical studies in mouse models of glioblastoma have demonstrated that HSV-SUR15 can effectively suppress tumor growth and establish long-term immune memory.
