HT-101 is a N-acetylgalactosamine (GalNAc)-conjugated small interfering RNA (siRNA) therapeutic developed for the treatment of chronic hepatitis B virus (HBV) infection. It works by targeting HBV RNAs for degradation via RNA interference, leading to reduced expression of viral proteins and pregenomic RNA. Preclinical and early clinical data show that subcutaneous administration of HT-101 results in significant reductions in key HBV biomarkers such as HBsAg, HBeAg, and HBV DNA across multiple genotypes. The drug has demonstrated safety and tolerability in phase I trials with healthy volunteers and patients with chronic hepatitis B, with higher doses associated with greater antigen reduction. The ongoing studies support its continued development as a promising strategy toward functional cure of chronic HBV infection[1][2][5].
