HT-K2 is a discovery-stage therapeutic candidate being developed by Hoth Therapeutics for the treatment of oncology and immune-related diseases. It is an antisense oligonucleotide (ASO) designed to target the K2 protein, also known as Kinesin-2 (specifically the KIF3A subunit). Kinesin-2 is a motor protein complex involved in intracellular transport and the formation of primary cilia, which are essential for various cell signaling pathways, including Hedgehog and Wnt. Dysregulation of KIF3A is associated with tumor progression, metastasis, and immune evasion in various cancers. By inhibiting the expression of KIF3A, HT-K2 aims to disrupt these oncogenic pathways and modulate the immune response to enhance anti-tumor activity. The program was initiated through a research collaboration with North Carolina State University.
