Overview

HT016 is a potent small molecule molecular glue degrader designed to target and induce the degradation of Vav guanine nucleotide exchange factor 1 (VAV1). VAV1 is a critical signaling protein primarily expressed in hematopoietic cells, where it plays a central role in T-cell and B-cell activation and downstream immune responses. By degrading VAV1 with high potency (DC50 as low as 0.10 nM), HT016 aims to modulate aberrant immune activity in autoimmune conditions. Developed by Wuhan Hiteck Biological Pharma in collaboration with LangRui Bio, the drug has shown efficacy in preclinical mouse models of rheumatoid arthritis (CIA), multiple sclerosis (EAE), and inflammatory bowel disease (IBD). It is currently in the pre-IND stage of development.
