hTAB004-CD3 is a preclinical-stage bispecific antibody T-cell engager being developed by OncoTab for the treatment of solid tumors, specifically triple-negative breast cancer (TNBC) and pancreatic cancer. It is derived from OncoTab's humanized monoclonal antibody, hTAB004, which selectively targets a tumor-associated form of MUC1 (tMUC1) that is aberrantly glycosylated and overexpressed in various cancers. The bispecific construct simultaneously binds to tMUC1 on the surface of cancer cells and the CD3 epsilon subunit of the T-cell receptor complex on T cells. This dual binding facilitates the formation of an immunological synapse, leading to the activation and redirection of T-cell-mediated cytotoxic activity against tMUC1-positive tumor cells, independent of MHC restriction. The development of hTAB004-CD3 has been supported by funding from the National Cancer Institute (NCI).
