HTI-101 is a recombinant human enzyme developed by Halozyme Therapeutics as part of its proprietary hyaluronidase platform. It was designed as a second patented enzyme in the company's portfolio, distinct from its lead rHuPH20 (Hylenex) technology. HTI-101 was primarily investigated for its potential therapeutic applications in oncology, inflammation, and lysosomal storage diseases. The drug's mechanism of action involves the enzymatic degradation of hyaluronic acid (hyaluronan), a major glycosaminoglycan component of the extracellular matrix. By breaking down hyaluronan, HTI-101 was intended to reduce interstitial fluid pressure within tumors to enhance the delivery of co-administered chemotherapeutic agents or to address pathological accumulations of hyaluronan in inflammatory conditions. The program was discontinued during the pre-clinical or early research phase and is no longer part of Halozyme's active development pipeline.
