HTI-1090 (also known as SHR9146) is an orally available small molecule that acts as a dual inhibitor of indoleamine 2,3-dioxygenase 1 (IDO1) and tryptophan 2,3-dioxygenase (TDO). By inhibiting these enzymes involved in the kynurenine pathway of tryptophan metabolism, HTI-1090 has potential immunomodulatory and antineoplastic effects. The drug is being developed primarily for the treatment of advanced solid tumors. IDO1 and TDO are implicated in tumor immune evasion by depleting tryptophan and producing immunosuppressive metabolites; their inhibition may restore anti-tumor immune responses. The drug was originally developed by Jiangsu Hengrui Medicine Co., with development also involving Atridia/Hengrui Therapeutics[1][2][4][5].
