HTI-1511 is an antibody-drug conjugate (ADC) targeting the epidermal growth factor receptor (EGFR), developed by Halozyme Therapeutics. It consists of a monoclonal antibody engineered for enhanced specificity within the tumor microenvironment, conjugated to the potent microtubule-disrupting cytotoxin monomethyl auristatin E (MMAE) via a novel bis-alkylating linker. This design aims to reduce the adverse skin reactions typically associated with EGFR-targeted therapies by minimizing interaction with receptors in normal tissue. Preclinical studies have demonstrated significant anti-tumor activity across various solid tumors, including head and neck squamous cell carcinoma, non-small cell lung cancer, and pancreatic carcinoma, even in models harboring downstream KRAS or BRAF mutations that often confer resistance to standard EGFR inhibitors.
