HTI-501 is an experimental recombinant human hyaluronidase variant developed by Halozyme Therapeutics. It was designed to target and degrade hyaluronan (hyaluronic acid), a major glycosaminoglycan component of the extracellular matrix that often accumulates at sites of chronic inflammation and edema. By enzymatically hydrolyzing hyaluronan, HTI-501 aimed to reduce interstitial fluid pressure, improve tissue permeability, and modulate the inflammatory microenvironment. While Halozyme's primary commercial success involves the use of rHuPH20 (Enhanze) to facilitate the subcutaneous delivery of other therapeutics, HTI-501 was specifically investigated for systemic administration to treat autoimmune and inflammatory diseases. The program was discontinued during early-stage development as the company shifted its focus toward oncology and its drug-delivery partnership platform.
