Overview

HTL0009936 is a potent and selective small molecule agonist of the M1 muscarinic acetylcholine receptor (M1 AChR), developed for the symptomatic treatment of cognitive dysfunction in Alzheimer's disease and other dementias. It exhibits high selectivity for M1 over other muscarinic receptor subtypes (no detectable activity at M2 and M3; seven-fold selectivity over M4). The drug has been studied in both oral and intravenous formulations, with clinical trials focusing on safety, pharmacokinetics, and exploratory pharmacodynamics in elderly subjects with below average cognitive functioning. Early studies indicate it is generally well-tolerated with dose-proportional pharmacokinetics but low oral bioavailability. Development aims to address cognitive symptoms associated with neurodegenerative diseases such as Alzheimer's disease[1][2][3][4][5][7].
