HTL0011076 (also known as SLIM-1) is a small molecule, orally available, selective agonist of the glucagon-like peptide-1 (GLP-1) receptor. Developed by Sosei Heptares (now Nxera Pharma) using its proprietary G protein-coupled receptor (GPCR) Structure-Based Drug Design (SBDD) platform, the compound was intended for the treatment of type 2 diabetes and obesity. HTL0011076 was designed to mimic the effects of endogenous GLP-1, which stimulates insulin secretion, inhibits glucagon release, and slows gastric emptying. However, development was discontinued in 2018 following a review of Phase 1 clinical trial data, which suggested that the compound's profile did not meet the criteria for further progression in a highly competitive market.
