HTL0027477 is a highly selective, first-in-class, oral small molecule agonist of G protein-coupled receptor 35 (GPR35), developed using Sosei Heptares' proprietary structure-based drug design platform. It is being investigated as a potential new treatment for inflammatory bowel diseases (IBD), including ulcerative colitis and irritable bowel syndrome. The drug has demonstrated promising preclinical efficacy and safety data, suggesting it may improve intestinal barrier function and reduce visceral pain in gastrointestinal disorders. Originally licensed to GSK in 2020, the rights were returned to Sosei Heptares in early 2024 after GSK deprioritized its immunology research strategy; this decision was not based on any scientific or safety concerns regarding the compound. Regulatory approval was granted by the UK MHRA in mid-2023 to commence first-in-human studies[2][3][4][5].
