HTL0048149 (also known as HTL'149) is a first-in-class, orally available small molecule drug developed by Sosei Heptares for the treatment of schizophrenia. It acts as a selective agonist of the orphan G protein-coupled receptor 52 (GPR52), representing a novel mechanism of action distinct from existing antipsychotic drugs. The drug is designed to address positive symptoms (such as psychosis, delusions, and hallucinations), negative symptoms (such as social withdrawal and apathy), and cognitive impairment (including deficits in attention, working memory, and executive function) associated with schizophrenia. Unlike current antipsychotics that often cause significant side effects and do not effectively treat negative or cognitive symptoms, HTL0048149 aims to provide an improved safety profile while targeting unmet needs in schizophrenia therapy. The compound was developed using Sosei Heptares’ StaR technology platform for structure-based drug design[1][3][5][6][7].
