hTLX3 (human T-cell leukemia homeobox 3) is a transcription factor and homeodomain protein that acts as a key oncogenic driver in a subset of T-cell acute lymphoblastic leukemia (T-ALL). In research settings, hTLX3 is typically delivered via retroviral or lentiviral vectors to hematopoietic stem cells or thymocytes to study its role in leukemogenesis. Overexpression of hTLX3, often in synergy with other mutations such as IL7Rα or NOTCH1, leads to an arrest in T-cell development and the promotion of cytokine-independent cell growth. It is a member of the TLX family, which includes TLX1 (HOX11), and is primarily associated with pediatric T-ALL cases.
