HTMC0435 + temozolomide is a combination therapy of an investigational oral PARP (poly(ADP-ribose) polymerase) inhibitor, HTMC0435, with temozolomide, an approved oral alkylating agent. HTMC0435 acts by inhibiting PARP enzymes involved in DNA repair, thereby enhancing the cytotoxicity of DNA-damaging agents such as temozolomide. Temozolomide is a methylating agent; its cytotoxic effects result from methylation of DNA, particularly at the guanine base, causing DNA damage and apoptosis when repair is inadequate. This combination has shown promising anti-tumor activity and a manageable safety profile in platinum-sensitive and platinum-resistant extensive-stage small cell lung cancer (ES-SCLC) in early clinical trials. Development is led by companies in China, with multiple phase 2 trials ongoing in solid tumors and pancreatic cancer[1][3][5].
