hTROP2-circRNA-LNP is an investigational cancer vaccine consisting of circular RNA (circRNA) encoding the human trophoblast cell surface antigen 2 (hTROP2) protein, encapsulated within lipid nanoparticles (LNPs). Developed by Orna Therapeutics, this vaccine platform utilizes circular RNA to provide enhanced stability and prolonged protein expression compared to traditional linear mRNA. hTROP2 is a transmembrane glycoprotein overexpressed in various epithelial cancers, making it a significant tumor-associated antigen for targeted immunotherapy. The vaccine is designed to stimulate a robust cellular immune response, specifically activating cytotoxic CD8α+ and helper CD4+ T cells against TROP2-positive tumor cells. Preclinical studies have demonstrated that its anti-tumor efficacy is significantly enhanced when co-administered with an IL-7-expressing circRNA-LNP, which promotes T-cell expansion and survival, helping to overcome self-tolerance and improve tumor infiltration.
