The **hTROP2 vaccine** is an investigational cancer immunotherapy targeting human TROP2 (trophoblast cell-surface antigen 2), a cell surface protein overexpressed in various solid tumors. It functions by eliciting a targeted immune response, stimulating tumor-infiltrating lymphocytes (TILs) that maintain an activated effector phenotype while modulating the tumor microenvironment (TME) by suppressing regulatory T cells (Tregs), myeloid-derived suppressor cells (MDSCs), and immunosuppressive cytokines such as IL-10 and TGF-β. Preclinical studies by CK Life Sciences using circular mRNA technology demonstrated 100% tumor growth inhibition in four of five mouse models, highlighting its potent anti-tumor efficacy. Additionally, related TROP2 vaccines like NIGel-Vax and AST-07X (Th1-specific) have shown up to 96% tumor suppression and 50-57% growth inhibition in triple-negative breast cancer (TNBC) and other models.[1][2][7][12]
