Overview

HTX-201 (formerly known as KHK-2455) is a small molecule inhibitor of indoleamine-pyrrole 2,3-dioxygenase 1 (IDO1), developed for the treatment and prevention of Epstein-Barr virus (EBV)-driven diseases. The drug has demonstrated preclinical efficacy in reducing EBV latency, tumor growth, and improving survival in models of post-transplantation lymphoproliferative disease (PTLD), a severe complication affecting high-risk solid organ transplant recipients. HTX-201 is being advanced by Hornet Therapeutics under an exclusive license from Kyowa Kirin. The company plans to initiate phase 1/2 proof-of-concept clinical trials in solid organ transplant populations within the next year. Future development may target other EBV-associated conditions such as multiple sclerosis, infectious mononucleosis, and long-COVID[1][2][3][4][8].
