Hu-alphaD11 is a humanized monoclonal antibody designed to target and neutralize Nerve Growth Factor (NGF), a neurotrophin that plays a critical role in the signaling and sensitization of pain. By binding to circulating NGF with high affinity, Hu-alphaD11 prevents the ligand from interacting with its high-affinity receptor, Tropomyosin receptor kinase A (TrkA), and its low-affinity receptor, p75 neurotrophin receptor (p75NTR), both of which are expressed on nociceptive neurons. This blockade effectively inhibits the peripheral sensitization of nociceptors and the subsequent transmission of pain signals. Originally derived from the murine antibody alphaD11 by Lay Line Genomics and humanized by PanGenetics, the candidate was later acquired by Abbott Laboratories (now AbbVie). Clinical development focused on chronic pain conditions such as osteoarthritis and interstitial cystitis; however, development was ultimately discontinued, following a trend in the anti-NGF class due to regulatory concerns regarding rapidly progressive osteoarthritis and other joint-related safety issues.
