HU1084-MMAE is an **antibody-drug conjugate (ADC)** consisting of a humanized anti-tissue factor monoclonal antibody (clone 1084, hu1084 mAb) conjugated to the potent cytotoxic agent **monomethyl auristatin E (MMAE)** via a protease-cleavable linker. It is designed to target tissue factor (TF/CD142)-expressing cancer cells, particularly in pancreatic cancer. The ADC binds to TF on the cancer cell surface, is internalized, and, upon lysosomal degradation, MMAE is released intracellularly to inhibit tubulin polymerization and induce cell cycle arrest and apoptosis. This targeted approach aims to enhance antitumor efficacy and reduce systemic toxicity associated with MMAE. Preclinical studies demonstrate strong antitumor effects in pancreatic cancer models, with efficacy dependent on TF expression levels[1].
