Overview

hu1B7 is a humanized monoclonal antibody designed to neutralize the pertussis toxin (PTx), a primary virulence factor of *Bordetella pertussis*. Developed by Synthetic Biologics (now Theriva Biologics) in collaboration with the University of Texas at Austin and Intrexon, hu1B7 is intended for the prophylaxis and treatment of pertussis (whooping cough), particularly in vulnerable infants. The antibody works by binding to the toxin and preventing its entry into host cells, thereby mitigating the severe leukocytosis and respiratory symptoms characteristic of the infection. It is a key component of the combination therapy SYN-005, which typically includes another antibody (hu3F11) to provide synergistic protection. Preclinical studies in non-human primates, supported by the Bill & Melinda Gates Foundation, have demonstrated its potential to prevent disease symptoms when administered as a passive immunization at birth.
