hu1G8-B is a **humanized monoclonal antibody** engineered to target **prostate stem cell antigen (PSCA)**, which is overexpressed in the majority of prostate cancers. It is derived by grafting the complementarity determining regions (CDRs) of the murine antibody 1G8 onto a human antibody framework, with additional murine residues in the CDR-H2 region to optimize antigen binding. Compared to its close relative hu1G8-A, hu1G8-B has more murine residues in key regions, resulting in a six-fold increase in binding affinity for PSCA. hu1G8-B showed the ability to bind PSCA with high specificity, inhibit tumor growth in vivo, and possess a unique cleft-like antigen binding site due to unusual loop truncations. Preclinical efficacy has been demonstrated primarily in prostate and bladder carcinoma models[1].
