Hu2F7 is a humanized monoclonal antibody (clone 2F7) that targets B7-H4 (also known as VTCN1), a member of the B7 family of immune checkpoint proteins. B7-H4 is frequently overexpressed in various epithelial malignancies, including breast, ovarian, and endometrial cancers, where it functions to suppress T-cell-mediated immune responses. Hu2F7 is widely utilized in the pharmaceutical industry as a benchmark or reference molecule for evaluating the performance of novel B7-H4-targeted therapies, such as biparatopic antibodies and antibody-drug conjugates (ADCs). When used in ADC formats, Hu2F7 facilitates the internalization of cytotoxic payloads into B7-H4-expressing tumor cells, leading to targeted cell death. Preclinical studies have demonstrated its efficacy in reducing tumor growth in various patient-derived xenograft (PDX) models, often serving as a comparative standard for next-generation B7-H4 targeting agents.
