Hu2G10 ADC (also known as LCB84) is an investigational antibody-drug conjugate (ADC) targeting the tumor-associated antigen Trop-2 (specifically the ADAM10-cleaved, activated form of Trop-2). It is composed of the humanized IgG1 monoclonal antibody Hu2G10 conjugated to the potent microtubule-disrupting agent monomethyl auristatin E (MMAE) via a plasma-stable, β-glucuronidase-cleavable linker. Developed using ConjuAll site-directed conjugation technology, Hu2G10 ADC is designed to selectively deliver its cytotoxic payload to cancer cells while sparing normal tissues that express wild-type Trop-2, thereby improving the therapeutic index. It is currently being evaluated in Phase 1/2 clinical trials for the treatment of various advanced solid tumors, including triple-negative breast cancer, non-small cell lung cancer, and pancreatic cancer.
