hu4G4 is a novel humanized monoclonal IgG1 antibody that specifically targets B7-H3 (also known as CD276), a transmembrane protein and immune checkpoint molecule that is highly overexpressed in various malignancies, including glioblastoma multiforme (GBM), but has limited expression in normal tissues. Developed by Suzhou Bright Scistar Antibody Biotech, hu4G4 binds to B7-H3 with high affinity (Kd ≈ 0.99 nM). While the antibody itself can inhibit tumor cell proliferation and migration, it is primarily utilized as a targeting moiety for radionuclide drug conjugates (RDCs), specifically when labeled with Iodine-131 (131I-hu4G4). In preclinical glioblastoma models, 131I-hu4G4 has demonstrated the ability to induce immunogenic cell death (ICD), increase the infiltration of CD4+ and CD8+ T cells, and promote the polarization of tumor-associated macrophages from an immunosuppressive M2 phenotype to a pro-inflammatory M1 phenotype, effectively converting the "cold" tumor microenvironment into a "hot" one.
