Overview

hu6M024 is a **humanized monoclonal antibody** (IgG1) that serves as the antibody component of antibody-drug conjugates (ADCs), specifically those targeting the cell surface protein PTK7 (protein tyrosine kinase 7). It is most notably used as the antibody in cofetuzumab pelidotin (PF-06647020), where hu6M024 is conjugated to an auristatin-derived microtubule inhibitor payload (Aur0101) via a cleavable valine-citrulline linker. The primary mechanism of action involves the binding of hu6M024 to PTK7 on cancer cells, subsequent internalization of the ADC, and intracellular release of the cytotoxic auristatin payload that inhibits microtubule formation, resulting in cell death. The major developer of the ADC utilizing hu6M024 is Pfizer. The antibody has been studied, mainly as part of the ADC, for use in solid tumors including ovarian cancer, non-small cell lung cancer, and triple-negative breast cancer[1][3][7][9].
