Overview

hu8E85 CART is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target Claudin 18.2 (CLDN18.2), a tight junction protein frequently overexpressed in gastrointestinal malignancies, including gastric, pancreatic, and esophageal cancers. The construct utilizes a humanized single-chain variable fragment (scFv) derived from the 8E85 antibody clone to recognize CLDN18.2. In preclinical studies, hu8E85 CART has demonstrated potent antitumor activity against CLDN18.2-positive xenografts; however, it has also been associated with significant off-tumor toxicity in healthy tissues expressing low levels of the target. Consequently, it is often utilized as a benchmark or comparator in the development of next-generation CAR-T therapies, such as nanobody-based or logic-gated (synNotch) systems, which aim to improve specificity and reduce adverse effects in the tumor microenvironment.
