Hu8F4 is a first-in-class, T cell receptor (TCR)-like human monoclonal antibody that specifically targets the leukemia-associated antigen PR1 presented by HLA-A2 (PR1/HLA-A2 epitope). By mimicking TCR specificity, Hu8F4 binds with high affinity to this peptide-MHC complex on the surface of malignant hematopoietic cells. The antibody has been studied both as a direct therapeutic agent and as the targeting domain in chimeric antigen receptor (CAR) constructs for adoptive cell therapy. Preclinical studies have demonstrated that Hu8F4 and its CAR-T derivatives can selectively recognize and kill PR1/HLA-A2-expressing leukemia cells. The primary clinical development focus is for advanced hematologic malignancies, including acute myeloid leukemia (AML), myelodysplastic syndromes (MDS), and other blood cancers[1][5][6].
