huA33 is a humanized monoclonal antibody that specifically targets the A33 antigen, a cell surface glycoprotein highly expressed in more than 95% of colorectal cancers and in normal intestinal epithelium but not in most other normal tissues. The mechanism of action involves binding to the A33 antigen on tumor cells, enabling selective targeting for imaging or therapeutic delivery—including radioimmunotherapy with radiolabeled forms such as 131I-huA33 or 124I-huA33. This targeted approach allows for high retention within tumors and rapid clearance from normal colonocytes, minimizing toxicity to healthy tissue. huA33 has been investigated primarily for metastatic colorectal cancer and also explored in gastric cancer expressing the A33 antigen[1][2][4][5][7].
