HuABC2 is a humanized IgG1 kappa monoclonal antibody antagonist targeting CD122, also known as the interleukin-2 receptor subunit beta (IL-2Rβ). Developed by JN Biosciences, the antibody is designed to treat immune-mediated diseases and prevent transplant rejection by blocking the signaling of both interleukin-2 (IL-2) and interleukin-15 (IL-15). HuABC2 possesses a unique mechanism of action that selectively suppresses CD122/CD132-expressing natural killer (NK) cells and CD8+ memory T cells, including tissue-resident memory T cells, while sparing CD25/CD122/CD132-expressing regulatory T cells (Tregs). This selective inhibition shifts the immune response toward tolerance. Preclinical studies in non-human primates have demonstrated that HuABC2 is well-tolerated, successfully suppresses the renewal of effector cells, and can prolong the survival of kidney allografts.
