Overview

huCART-19 is a **humanized CD19-targeted chimeric antigen receptor (CAR) T-cell therapy** designed to treat B-cell malignancies, especially relapsed or refractory B-cell acute lymphoblastic leukemia (B-ALL) and B-cell lymphomas. This therapy involves collecting a patient's own T cells and genetically modifying them to express a CAR that specifically targets CD19, a protein found on the surface of B cells. The modified T cells are expanded and re-infused into the patient, where they seek out and destroy malignant B cells. huCART-19 addresses challenges in CAR T-cell persistence and relapse by utilizing a humanized antibody fragment in its CAR design, potentially reducing immune rejection and allowing for longer T-cell survival. More recently, an "armored" version, huCART19-IL18, has been developed, where the cells are engineered to also secrete interleukin-18 (IL-18), a pro-inflammatory cytokine, enhancing antitumor activity and recruiting additional immune support[2][4][5]. The therapy is primarily studied in patients who have failed previous CAR T or other therapies[1][2][4].
