Drug intelligence / Profile preview
`huCART19-IL18-CD428z` is an experimental, fourth-generation, "armored" chimeric antigen receptor (CAR) T-cell therapy developed by the University of Pennsylvania. It is designed to treat patients living with HIV who have relapsed or refractory CD19-positive B-cell malignancies, including non-Hodgkin lymphoma (NHL), chronic lymphocytic leukemia (CLL), acute lymphoblastic leukemia (ALL), and Richter's transformation. The therapy consists of autologous T cells genetically engineered via co-transduction with two lentiviral vectors: one encoding a humanized anti-CD19 CAR that constitutively secretes the pro-inflammatory cytokine interleukin-18 (IL-18) to enhance antitumor activity and modulate the tumor microenvironment, and another encoding a C34-CD4CAR_CD28 construct targeting HIV. By co-expressing CD19-targeting and HIV-specific CARs, the therapy aims to leverage CD19-positive cancer cells to stimulate T-cell proliferation and survival, thereby enhancing the persistence of the HIV-specific CAR T cells to help control the viral reservoir. It is currently being evaluated in a Phase 1 clinical trial.
