Overview

huCD19.28.z-CAR is a second-generation chimeric antigen receptor (CAR) construct designed to target the human CD19 antigen. The receptor typically consists of a humanized or human-derived single-chain variable fragment (scFv) specific for CD19, linked to a CD28 costimulatory domain and a CD3-zeta (CD3ζ) signaling domain. While CD19-targeted CAR-T therapies have been highly successful in treating hematological malignancies, this specific construct is utilized in research to investigate strategies for overcoming the challenges of solid tumors, such as the immunosuppressive tumor microenvironment and heterogeneous antigen expression. In preclinical studies, it often serves as a benchmark control for next-generation CARs, such as those engineered to secrete cytokines like IL-18 to recruit endogenous immune cells and enhance anti-tumor efficacy against antigen-negative cells.
