huHER3-8

huHER3-8 is a humanized monoclonal antibody designed to target the ErbB3 (HER3) receptor. Developed by ImmunoGen, the antibody functions by binding to the extracellular domain of ErbB3, thereby neutralizing its activity and preventing ligand-induced (e.g., neuregulin-1) activation. This inhibition disrupts the formation of ErbB3/ErbB2 heterodimers and suppresses downstream signaling pathways, most notably the PI3K/AKT pathway, which is critical for tumor cell survival and proliferation. Preclinical research has demonstrated its potential in treating subsets of melanoma, particularly those with wild-type BRAF and NRAS that exhibit high basal ErbB3 signaling driven by autocrine neuregulin-1 production.
