**huIgG1-sulfo-SPDB-DM4** is a nontargeting human IgG1 antibody-drug conjugate used as an experimental isotype-control ADC in preclinical studies of folate receptor alpha-targeted ADCs. It consists of a human IgG1 antibody conjugated through a hydrophilic, hindered disulfide sulfo-SPDB linker to the maytansinoid payload DM4. Unlike IMGN853, this construct does not bind folate receptor alpha and was used to demonstrate that antitumor activity of the targeted ADC was antigen dependent. Intracellular release of DM4 can inhibit microtubule function, causing mitotic arrest and cell death, but the construct itself was not developed as a clinical therapeutic product.
