huJ591 is a humanized monoclonal antibody that targets the extracellular domain of prostate-specific membrane antigen (PSMA), a cell surface peptidase highly expressed on malignant prostate epithelial cells and the vascular endothelium of various solid tumors. The antibody was developed by deimmunizing murine J591 to reduce immunogenicity in humans. huJ591 has been studied both as an unconjugated antibody and as a radioimmunoconjugate with beta-emitting (e.g., lutetium‑177 [^177Lu], yttrium‑90 [^90Y]) or alpha-emitting (e.g., actinium‑225 [^225Ac]) radionuclides for targeted radiotherapy. Clinical trials have demonstrated its ability to specifically target metastatic prostate cancer lesions and other PSMA-expressing tumors with favorable safety profiles. Its primary indication is metastatic castration-resistant prostate cancer (mCRPC), but it has also shown potential in other urological and solid tumor malignancies[2][4][5][6][9].
