Overview

huJCAR014 (also known as JCAR021) is an investigational, humanized CD19-targeted chimeric antigen receptor (CAR) T-cell therapy developed by the Fred Hutchinson Cancer Center. It is a second-generation CAR-T product consisting of a defined 1:1 ratio of CD4+ and CD8+ T cells. Unlike its predecessor JCAR014, which utilized a murine-derived single-chain variable fragment (scFv), huJCAR014 incorporates a fully human scFv to minimize immunogenicity and enhance the persistence of the CAR T cells in the patient's body. The construct includes a 4-1BB (CD137) costimulatory domain and a CD3-zeta signaling domain, along with a truncated epidermal growth factor receptor (EGFRt) as a safety switch or selection marker. It is primarily being evaluated for the treatment of relapsed or refractory B-cell malignancies, including B-cell non-Hodgkin lymphoma and acute lymphoblastic leukemia.
