HuLuc63-CAR-T is an investigational chimeric antigen receptor (CAR) T-cell therapy designed to target Signaling Lymphocytic Activation Molecule Family member 7 (SLAMF7), also known as CS1 or CRACC. SLAMF7 is a cell surface glycoprotein that is highly and uniformly expressed on multiple myeloma cells, making it a promising target for cellular immunotherapy. The CAR construct utilizes a humanized single-chain variable fragment (scFv) derived from the monoclonal antibody HuLuc63 (elotuzumab), which specifically binds to the proximal Ig-like constant 2 (C2) domain of SLAMF7. Upon binding to the target on myeloma cells, the engineered T cells are activated to exert potent cytotoxic effects. A notable challenge in the development of SLAMF7-targeted CAR-T therapies is 'fratricide' (self-killing), as SLAMF7 is also expressed on a subset of normal lymphocytes, including NK cells and CD8+ T cells. HuLuc63-CAR-T is primarily being developed for the treatment of relapsed or refractory multiple myeloma.
