Overview

Human amniotic epithelial cell-derived extracellular vesicles (hAEC-EVs) are a cell-free therapeutic modality being investigated for their regenerative, anti-inflammatory, and immunomodulatory properties. These vesicles are nano-sized, membrane-bound particles secreted by human amniotic epithelial cells, which are harvested from the innermost layer of the placenta. hAEC-EVs carry a complex cargo of bioactive molecules, including microRNAs (miRNAs), proteins, and lipids, that facilitate paracrine signaling to recipient cells. In the context of dry eye disease (DED), hAEC-EVs have demonstrated the ability to reduce ocular surface inflammation, promote the repair of the corneal epithelium, and restore tear film stability by suppressing oxidative stress and modulating pro-inflammatory signaling pathways. As a cell-free alternative to stem cell therapy, they offer advantages such as lower immunogenicity, easier storage, and a reduced risk of tumorigenicity.
