Overview

Human amniotic membrane allograft is a biologic tissue graft derived from the innermost lining of the placenta, specifically the amnion and chorion, collected after birth, cleaned, sterilized, and processed (either cryopreserved or dehydrated) for clinical use[4][7][12]. It contains collagen, fibronectin, hyaluronic acid, growth factors, cytokines, and anti-inflammatory proteins such as interleukin-1 receptor antagonist, providing anti-inflammatory, anti-fibroblastic, antimicrobial, analgesic, anti-angiogenic (or pro-angiogenic, depending on orientation), and immunomodulatory properties[12][11]. HAM allografts are nonimmunogenic and promote tissue regeneration, making them effective for a variety of clinical indications including non-healing diabetic ulcers, venous stasis ulcers, pressure sores, burns, ophthalmic surface disorders (corneal ulcers/melts, neurotrophic keratitis, bullous keratopathy, Stevens-Johnson syndrome, pterygium repair), surgical wound healing, skin injury, tendon repair, and as anti-adhesion barriers for surgical procedures[11][4][12][9]. The mechanism involves elution of growth factors, modulation of inflammation (downregulation of TNF-α, IL-1β, IL-8, TGF-β1), ECM modification, inhibition of fibrosis, and stimulation of epithelial and mesenchymal stem cell migration[11][12].
