Overview

Human epidermal growth factor receptor 2-chimeric antigen receptor cytomegalovirus-specific cytotoxic T lymphocytes (HER2-CAR CMV-CTLs) are an investigational cellular immunotherapy developed primarily at Baylor College of Medicine. This approach involves engineering T cells that are naturally specific for Cytomegalovirus (CMV) antigens to express a chimeric antigen receptor (CAR) that targets HER2 (ERBB2). The rationale behind using virus-specific T cells as a substrate for CAR engineering is to utilize the cells' native T-cell receptors (TCRs) to receive survival and proliferation signals from endogenous CMV antigens, which may enhance the persistence and expansion of the CAR-T cells in the immunosuppressive tumor microenvironment. This therapy has been primarily evaluated for the treatment of HER2-positive glioblastoma multiforme, where it aims to overcome the limited persistence typically observed with conventional CAR-T cell products.
