Human heavy chain ferritin (HFn) nanoparticles are bio-inspired, protein-based nanocages used as a targeted drug delivery platform. Composed of 24 self-assembling subunits of the human ferritin heavy chain, these nanoparticles naturally target the transferrin receptor 1 (TfR1/CD71), which is frequently overexpressed on the surface of various cancer cells, including acute myeloid leukemia (AML) blasts. Upon binding to CD71, HFn nanoparticles undergo receptor-mediated endocytosis, facilitating the intracellular delivery and release of encapsulated therapeutic agents. This system is particularly effective for delivering hydrophobic chemotherapies, such as venetoclax, by improving their solubility, enhancing tumor-specific accumulation, and potentially reducing systemic toxicity.
