human interleukin-2 messenger RNA + aNP-HMJ1 is an experimental cancer immunotherapy comprising messenger RNA (mRNA) encoding human interleukin-2 (hIL-2) encapsulated within apolipoprotein A1 nanoparticles (aNPs) that utilize the proprietary ionizable lipid HMJ1. Developed by Biotrip in collaboration with Eindhoven University of Technology and Radboud University Medical Center, the platform is designed for the targeted delivery of cytokine-encoding mRNA to secondary lymphoid organs, such as the spleen and lymph nodes, while minimizing off-target expression in the liver. This localized production of hIL-2 is intended to expand and activate cytotoxic CD8+ T cells, enhancing their anti-tumor effects without the systemic toxicities associated with traditional recombinant IL-2 therapy. Preclinical studies in melanoma and colorectal cancer models have demonstrated significant tumor growth inhibition and complete remissions, supported by biodistribution data in non-human primates.
