Human islet amyloid polypeptide (hIAPP) is a protein that forms pathogenic aggregates. These aggregates are known to damage pancreatic islet β cells in type 2 diabetes mellitus (T2DM) and are implicated in T2DM-associated peripheral neuropathy. Aggregated hIAPP is neurotoxic, reducing neurite outgrowth and increasing mitochondrial reactive oxygen species in sensory neurons. Its aggregation is a key factor in mediating peripheral neuropathy, and pharmacological inhibition of hIAPP aggregation has been shown to prevent these neuropathic effects.
