Overview

Human keratinocyte growth factor (KGF, also known as fibroblast growth factor 7 or FGF-7) is a member of the fibroblast growth factor family, produced endogenously by mesenchymal cells. Recombinant versions (often called rHuKGF) are manufactured using recombinant DNA technology. KGF binds to the keratinocyte growth factor receptor (a splice variant of the fibroblast growth factor receptor 2), which is found on epithelial cells but not on hematopoietic (blood cell) lineages. Binding of KGF to its receptor stimulates the proliferation, differentiation, and migration of epithelial cells, resulting in cytoprotection and regeneration of mucosal and epithelial surfaces following injury from chemotherapy or radiation. Recombinant human KGF, such as palifermin, is used clinically to reduce and shorten the duration of severe oral mucositis in patients receiving cytotoxic cancer therapy and is being investigated for broader epithelial protective effects, such as in acute respiratory distress syndrome and gastrointestinal mucositis[1][2][3][4][6][8].
