This next-generation prophylactic vaccine candidate for human papillomavirus (HPV) infection is being developed by the German Cancer Research Center (Deutsches Krebsforschungszentrum, DKFZ) in collaboration with the Helmholtz Centre for Infection Research (HZI). The vaccine combines a recombinant HPV L2 capsid protein antigen with the novel adjuvant cyclic di-AMP (c-di-AMP). Unlike current L1-based virus-like particle (VLP) vaccines that are largely type-specific, the L2 antigen targets a highly conserved region of the minor capsid protein (often incorporating the RG1 epitope), which aims to provide broad-spectrum, cross-neutralizing protection against a wide range of high-risk and low-risk HPV genotypes. The adjuvant c-di-AMP is a STING (Stimulator of Interferon Genes) agonist that significantly enhances both systemic and mucosal immune responses. This combination is particularly notable for its potential efficacy via alternative administration routes, such as intranasal delivery, which could simplify global vaccination efforts and improve mucosal immunity at the site of infection.
