Overview

The human papillomavirus (HPV) nanovaccine is an experimental therapeutic cancer vaccine developed by researchers at UT Southwestern Medical Center. It consists of 25-30 nm nanoparticles that encapsulate the HPV16 E7 protein antigen along with a dual-component adjuvant comprising a pH-responsive polymer and the small-molecule STING agonist di-amidobenzimidazole (diABZI). Designed for systemic administration, the nanovaccine is responsive to acidic environments and NAD(P)H quinone oxidoreductase 1 (NQO1), facilitating the delivery of its cargo to immune cells in the spleen. By activating the Stimulator of Interferon Genes (STING) pathway, the vaccine primes a robust T-cell response against HPV-infected tumor cells, demonstrating the ability to eradicate both primary tumors and metastatic nodules in preclinical models of cervical and head and neck cancers.
