Overview

Human papillomavirus type 16 L1 protein is a recombinant antigen that self-assembles into virus-like particles (VLPs), which are morphologically and antigenically similar to the native HPV 16 virion but lack the viral genome. Developed by CHU Saint Pierre, this protein-based vaccine candidate is designed to elicit a robust humoral immune response, specifically inducing high-titer neutralizing antibodies against the L1 capsid protein. These antibodies prevent the virus from binding to and entering host epithelial cells, thereby providing prophylaxis against HPV 16 infection. This high-risk HPV genotype is the primary cause of cervical cancer and is strongly associated with other malignancies and precursor lesions, including cervical, vulvar, and vaginal intraepithelial neoplasia (CIN, VIN, and VaIN). Clinical development has focused on evaluating the safety and immunogenicity of this VLP-based approach in various populations, including those with increased vulnerability to persistent HPV infection.
