Overview

Human papillomavirus type 16 L1 protein (AS04-adjuvanted) is a monovalent prophylactic vaccine candidate designed to prevent infection by HPV type 16, a high-risk genotype primarily responsible for cervical cancer and other anogenital malignancies. The vaccine is composed of recombinant L1 capsid proteins that self-assemble into virus-like particles (VLPs), which are non-infectious but highly immunogenic due to their structural similarity to the native virion. It utilizes the proprietary AS04 adjuvant system, which combines aluminum hydroxide with 3-O-desacyl-4'-monophosphoryl lipid A (MPL), a Toll-like receptor 4 (TLR4) agonist. This combination is intended to enhance the magnitude and duration of the humoral immune response compared to aluminum salts alone. While this monovalent formulation was extensively evaluated in early clinical trials, it served as the technological precursor to the bivalent vaccine Cervarix (HPV-16/18). The Geneeskundige En Gezondheidsdienst (GGD) Amsterdam has been a significant clinical research partner in the long-term evaluation of this vaccine's immunogenicity and efficacy.
